Functional analysis in the KIF7 gene expression

Intervention(s)I gather 14 mL blood of a patient of the PAC syndrome, and a control, in the study purpose concerned newly. I measure serum IL-1 beta, IL-6, TNF-alpha density by ELISA. I isolate PBMC. 1)It is stimulation with TLR ligand(TL4, TLR9, TLR1/2, TLR2, TLR2/6, TLR3, TLR5, TLR7, TLR8). 2) I gather supernatant liquid of PBMC which I stimulated, and PBMC not to stimulate, and measure IL-1 beta, IL-6, TNF-alpha density. 3) I separate RNA from PBMC which I stimulated, and PBMC not to stimulate. I reverse-transcript it to cDNA and, using molds such as KIF7, perform rtPCR. I make SiRNA of the KIF7-related factor and introduce a gene into PBMC by transfection and check a change including the expression of KIF7.

Include criteriaobject person(patient) In Kurume University Hospital collagenosis internal medicine, I intend for a patient under treatment for PAC syndrome.
